If you are going by the exact phrasing of the question, then yes, the Infinity Gauntlet is the most powerful item in comics. The Heart of the Universe isn't an actual item.

Having a hard time remembering what the Heart actually looked like in the comic. I think it was like a big ball of energy that Thanos stuck his hand into merging the Heart within himself. It gave him omnipotent cosmic powers. He absorbs Living Tribunal, Eternity, Infinity and Galactus along with the entire universe into himself and is left all alone in darkness.
