I am not a spiderman expert but I thought MJ and Blackcat were Spiderman’s love interest and never to each other? Ivy and Harley are an actual romantic couple and has been confirmed by DC and they even had them married in one of the universe...How are they just suggestive theme?

I am just gonna think you are not up to date with comics. Both marvel and DC have gay couples. DC most notably Poison Ivy/Harley, Batwoman/Question, Apollo/Midnighter. Marvel has Wiccan/Hulkling, Destiny/Mystique from what I remember. So no, I don’t agree that this is a first.

How is Harley and Ivy being a couple a "subjective theme"? The two have had an implied romantic relationship for almost as long as Harley has existed, and nowadays DC allows them to be openly depicted as lovers in the comics. Despite claiming not to have a problem with that depiction of the two, it kind of seems like you do.

How is this remotely "a first?" They couldn't depict them as being "together" more unless they were full on scissoring in the comics.

And like chrismhk mentioned, there's a long history of both companies having same sex couples. This isn't a first by any means.
