Crisis on Infinite Earths. To this day, when talking about DC continuity or history, you still say Pre-Crisis or Post-Crisis.

Besides the release of a character (obviously Action Comics #1 would be first) I'd say that it would be Superman #76, this is the first Batman/Superman team up and introduced the concept a universe.

Fantastic Four No. 1: Began the Marvel Era of comics, which took the industry to new heights. There is not a single more imporant creator in comics history than Stan Lee, and FF 1 is where he began to create his universe.

Good point! But I think if a SINGLE most important comics creator is to be chosen, it has to be Stan. Lee, Kirby and Ditko were Marvel's main three creative forces early, but Stan guided the entire company from a great start to continued greatness through the 1980s. Nobody tops The Man!
